Theoleptus II of Constantinople served as Ecumenical Patriarch from 1585 to 1586. A nephew of Patriarch Metrophanes III, he was appointed Metropolitan of Philippopolis before conspiring against Patriarch Jeremias II with Pachomius II. When Pachomius II was deposed in February 1585, Theoleptus replaced him and was enthroned by the Patriarchs of Alexandria and Antioch in March 1585. His tenure ended abruptly when Nicephorus, a deacon loyal to Jeremias II, dethroned him in May 1586 while he was fundraising in Moldavia and Wallachia. Nicephorus acted as locum tenens until April 1587, when Jeremias II was re-elected despite his absence on a journey to Ukraine and Russia. Jeremias II did not return to Constantinople until 1590.

Theoleptus II was later pardoned by a synodal decision after April 1587 and sent to Iberia to raise funds for the indebted Church. In 1587, he issued a signet for the Soumela Monastery, though its legality remains unclear. Theoleptus eventually reconciled with Jeremias II and assisted in governing the Church until 1590. His fate after that is unknown.

Theoleptus II was the last Patriarch to have his see at the Pammakaristos Church, which was converted into a mosque in 1586. The Patriarchate then moved to the poorer Church of Theotokos Paramythia, where it remained until 1597.
